The human arginase-I kit has been developed for the quantitative measurement of human arginase-I in cell culture medium, serum and plasma of both natural and recombinant origin. Arginase (L-arginine ureahydrolase, or L-arginine amidinohydrolase) is a hydrolytic enzyme that converts arginine to ornithine and urea. Two forms of arginase exist which are designated as arginase-I and arginase-II. Although arginase-I and arginase-II have similar enzyme activities, they have different pI and immunological reactivity. They are encoded by different genes. Human arginase-I is a 35 kD protein circulating in blood probably as a homotrimer. Liver-type arginase-I is most abundant in mammalian liver, but it is also found in non-hepatic tissues, such as red blood cells, lactating mammalian glands, and the kidney. In addition to its involvement in ammonia detoxification via the urea cycle, arginase plays a role in other processes, such as macrophage-mediated cytotoxicity due to arginase release and inhibition of lymphocyte proliferation in vitro. It shows high activity in growing tissues, proliferating lymphocytes and tumors. Furthermore, arginase acts as a modulator of the immune response. Besides, arginase plays a role in allergen challenged lungs and in autoimmune inflammation in the central nervous system. |
