Description:

Both MIP-1α and MIP-1β are structurally and functionally related CC chemokines. They participate in the host response to invading bacterial, viral, parasite and fungal pathogens by regulating the trafficking and activation state of selected subgroups of inflammatory cells e.g. macrophages, lymphocytes and NK cells. While both MIP-1α and MIP-1β exert similar effects on monocytes their effect on lymphocytes differ; with MIP-1α selectively attracting CD8+ lymphocytes and MIP-1β selectively attracting CD4+ lymphocytes. Additionally, MIP-1α and MIP-1β have also been shown to be potent chemoattractants for B cells, eosinophils and dendritic cells. Both human and murine MIP-1α and MIP-1β are active on human and murine hematopoietic cells. Recombinant human MIP-1α is a 7.8 kDa protein containing 69 amino acid residues, including the four highly conserved cysteine residues present in CC chemokines.

Reconstitution:

Centrifuge vial prior to opening. Reconstitute in sterile ddH2O to a concentration of 1 mg/ml. The solution can then be further diluted in other aqueous buffers.

Application notes:

ELISA:  In a sandwich ELISA (assuming 100 µl/well), a concentration of 2 -4µg/ml of this antibody will detect at least 2000 pg/ml of recombinant human MIP-1α when used with biotinylated antigen affinity purified anti-human MIP-1α as the detection antibody at a concentration of approximately 1 -2 µg/ml.
Western Blot:  To detect Human MIP-1α by Western Blot analysis this antibody can be used at a concentration of 2 - 4 µg/ml. When used in conjunction with compatible secondary reagents, the detection limit for recombinant Human MIP-1α is 1 - 2 ng/lane, under nonreducing conditions.
Neutralization:  To yield one-half maximal inhibition [ND50] of the biological activity of Human MIP-1α (100 ng/ml), a concentration of 5 - 10 μg/ml of this antibody is required.
The optimal concentration should be determined by the user for each application.
